If a fanatic blocks a player on his own team, can that player use defensive assists against the fanatic?

you would probably be better off not using defensive assists

if you don't then the fanatic rolls 3 dice, and with 3 dice there is the highest chance of getting a pushback

less dice you roll the less options you will have to choose from

Right, that makes sense, dumb question I guess. But since we´re on the subject, are you allowed to use assists against your own team mates, theoretically speaking?

yes, as the goblin coach, you can assign assists as you wish (and i'd give them to the fanatic to ensure he gets 3 dice)

what's less clear to me is whether the opponent can give assists to either the fanatic or the teammate

if you mean for normal blocks, i don't think so, but why would you do it anyway?
