I just want to be sure on a point: does the 2nd defender involved in a Multiple Block can assist his team-mate on the 1st block ?
I suppose that the answer is yes, so it means that the 2 defenders are no longer considered as a single entity like in LRB 4 right ?
That is correct, in LRB 5.0 two separate blocks are made each calculated separately. So make the first block (defender +2 ST) taking assists into account. Then make the second block (defender +2 ST) taking assists into account.
It is worth mentioning that the second block does not need to be adjacent to the first one.

Also note that you get to decide in which order the blocks take place, so if one defender can assist and the other can't, then you can use that to your advantage.

One short question here, is Dauntless/Multiple Block a valid combo?
Example: Blocker ST 3 M.Block & Dauntless vs. 2 Defenders ST3 (+2). Can you use Dauntless to equal their ST 5 or is the M.Block +2ST bonus for defenders applied after dauntless?

You would roll for Dauntless before each block and each time it would be against ST 5 as the modifiers are applied before Dauntless is rolled.
